The use of computed tomography (CT) scan is essential for making diagnoses for trauma patients in emergency medicine. Numerous studies have been conducted on guiding medical examinations in light of advances in machine learning, leading to more accurate and rapid diagnoses. The present study aims to propose a machine learning-based method to help emergency physicians prevent performance of unnecessary CT scans for chest trauma patients. A dataset of 1000 samples collected in nearly two years was used. Classification methods used for modeling included the support vector machine (SVM), logistic regression, Naïve Bayes, decision tree, multilayer perceptron (four hidden layers), random forest, and K nearest neighbor (KNN). The present work employs the decision tree approach (the most interpretable machine learning approach) as the final method. The accuracy of 7 machine learning algorithms was investigated. The decision tree algorithm was of higher accuracy than other algorithms. The optimal tree depth of 7 was chosen using the training data. The accuracy, sensitivity and specificity of the final model was calculated to be 99.91% (95%CI 99.10% - 100%), 100% (95%CI 99.89% - 100%), and 99.33% (95%CI 99.10% - 99.56%), respectively. Genetic TET2 deficiency leads to skewed myeloid differentiation and clonal expansion, but minimal residual TET activity is critical for survival of neoplastic progenitor and stem cells. Consistent with mutual exclusivity of TET2 and neomorphic IDH1/2 mutations, here we report that IDH1/2 mutant-derived 2-hydroxyglutarate is synthetically lethal to TET-dioxygenase deficient cells. In addition, a TET-selective small molecule inhibitor decreased cytosine hydroxymethylation and restricted clonal outgrowth of TET2 mutant, but not normal hematopoietic precursor cells in vitro and in vivo. We studied pretreatment and relapse bone marrow samples from patients with FLT3-mutated AML treated with FLT3i-based therapies (secondary resistance cohort), and pretreatment bone marrow samples from patients with no response to FLT3i-based therapies (primary resistance cohort). Targeted next generation sequencing at relapse identified emergent mutations involving on-target FLT3, epigenetic modifiers, RAS/MAPK pathway, and less frequently WT1, and TP53. RAS/MAPK and FLT3-D835 mutations emerged most commonly following type I and type II FLT3i-based therapies, respectively. Patients with emergent mutations at relapse had inferior overall survival compared with those without emergent mutations. Among pretreatment RAS mutated patients, pretreatment cohort level variant allelic frequencies for RAS were higher in non-responders, particularly with type I FLT3i-based therapies, suggesting a potential role in primary resistance as well. However, it is challenging for perovskite PVs due to the lack of eco-friendly solvents for ambient fast printing. In this study, we demonstrate for the first time an eco-friendly printing concept for high-performance perovskite solar cells. Both the perovskite and charge transport layers were fabricated from eco-friendly solvents via scalable fast blade coating under ambient conditions. The perovskite dynamic crystallization during blade coating investigated using in situ grazing incidence wide-angle X-ray scattering (GIWAXS) reveals a long sol-gel window prior to phase transformation and a strong interaction between the precursors and the eco-friendly solvents. The insights enable the achievement of high quality coatings for both the perovskite and charge transport layers by controlling film formation during scalable coating.
